History and Info
Pristiq (Desvenlafaxine) is the active metabolite of Effexor (Venlafaxine) and was approved in January 2007. While Pristiq was approved in both the United States and Canada, the European Union declined the status, stating in part that Pristiq (marketed under the name Ellefore in Europe) "the Committee for Medicinal Products for Human Use had some concerns and was of the provisional opinion that Ellefore could not have been approved for the treatment of major depressive disorder [and] overall, the effectiveness of Ellefore had not been shown convincingly. In relation to its parent substance, Venlafaxine, Desvenlafaxine seemed to be less effective with no advantages in terms of safety and tolerability." Wyeth subsequently withdrew its application for approval.

In December 2015 Pfizer announced the results from a Phase 3 study evaluating the efficacy, safety and tolerability of Pristiq (sustained release) in pediatric patients ages 7-17 with Major Depressive Disorder. The results indicate that Pristiq were not significantly different from placebo.

Pristiq has also shown higher rates of discontinuation syndrome (withdrawals) than other SSRI or SNRI antidepressants. The symptoms can be severe and considered “intolerable” by many patients and therefore the need to gradually taper is highly recommended.
